Introduction
Il y a presque 20 ans, Schabel rappelait que le criblage in vivo était la «pierre angulaire» du développement en chimiothérapie anticancéreuse [33]. Cette affirmation est, à notre avis, toujours d’actualité. En effet, en l’absence de cibles clairement identifiées et validées permettant une sélectivité antitumorale, les modèles de tumeurs in vivo représentant des pathologies cancéreuses variées sont aujourd’hui encore, l’outil majeur pour la détection et l’évaluation de nouvelles molécules antinéoplasiques [34] et une étape obligatoire avant les premières études cliniques (phase I). Récemment encore, le National Cancer Institute américain (NCI) rappelait qu’il ne pouvait y avoir de progrès en chimiothérapie anticancéreuse expérimentale sans l’utilisation de l’expérimentation animale [3]. Il faut aussi rappeler que l’activité de la quarantaine de molécules antinéoplasiques actuellement utilisées en clinique a été découverte et/ou évaluée sur des modèles murins.
Les tumeurs greffées chez l’animal ont l’avantage de présenter plusieurs caractéristiques de la pathologie humaine qu’elles cherchent à représenter. En effet, les animaux porteurs de tumeurs possèdent des barrières physiologiques limitant l’accès du produit à la tumeur, des organes assurant le métabolisme et l’excrétion du produit, des tissus sains qu’il faut sauvegarder, des métastases, et aussi toutes les cibles rationnelles de la maladie cancéreuse elle-même, avec son hétérogénéité et ses mécanismes de résistance.
Néanmoins, il faut insister sur le fait qu’il n’y a pas de modèle tumoral animal prédictif sans équivoque de la réponse aux médicaments anticancéreux chez l’homme (à l’exception peut-être de la leucémie L1210 pour la leucémie lymphoblastique aiguë) et donc la découverte de faux positifs par ces modèles est inévitable. Il serait utopique de penser qu’un seul modèle puisse être le reflet de toutes les tumeurs humaines si l’on tient compte de la diversité des cancers humains, chaque tumeur maligne étant une entité, avec un comportement biologique et une réponse aux produits, qui lui sont propres [20], sans oublier les interactions complexes tumeur-hôte et produits-tumeur-hôte. L’efficacité d’une molécule envers plusieurs tumeurs animales d’un même type histologique pourrait peut-être prédire plus sûrement une efficacité chez l’homme dans le même type de tumeur.
Pour mettre en perspective les problèmes actuels de dépistage de nouveaux produits antitumoraux, nous aborderons successivement l’historique des modèles expérimentaux utilisés en chimiothérapie anticancéreuse, les différentes stratégies actuelles de criblage, et enfin la méthodologie d’évaluation des agents antinéoplasiques in vivo, en insistant sur les tumeurs solides.
Historique du développement des premiers modèles tumoraux et de leur utilisation dans le criblage d’agents anticancéreux
La première transplantation tumorale chez l’animal est attribuée à Novinsky (1875) pour le passage d’une tumeur nasale et d’un myxosarcome vénérien chez le chien [40]. Vers la même époque, Wehr publie ses travaux sur la propagation de tumeurs génitales chez le chien (1888-1889), et Hanau transfère avec succès un carcinome vulvaire chez le rat [40].
Morau est le premier à étudier systématiquement les relations hôte-tumeur en utilisant un épithélioma de souris [28]. Par la suite, les travaux de Jensen sur la transplantation d’un carcinome alvéolaire spontané chez la souris blanche sont considérés comme le point de départ des travaux de recherche sur les tumeurs murines en Europe et aux Etats-Unis [25]. En effet, les tumeurs de Jensen, ainsi que d’autres tumeurs isolées par Ehrlich et Borrel sont obtenues et propagées avec succès dans plusieurs laboratoires, notamment ceux de Clowes [10] et de Tyzzer [41].
La nécessité de souris consanguines pour l’étude des cancers est reconnue dès le début des années 1900 par de nombreux chercheurs intéressés par les facteurs génétiques liés au développement des tumeurs mammaires. Ceux-ci contribuent au développement d’un certain nombre de souches murines: C57BL/6, C57BL/10, DBA, BALB/c (Little); C3H, CBA (Strong); AKR (Furth) [29]. C’est le développement de ces lignées de souris consanguines qui va permettre l’utilisation de tumeurs greffées encore utilisées aujourd’hui.
Le développement des méthodes de criblage de substances antitumorales est intimement lié au développement des modèles tumoraux chez l’animal. Ainsi le développement des sarcomes 37 et 180 (souris), du carcinosarcome 256 de Walker (rat), et du carcinome ascitique d’Ehrlich (développés avant 1930) vont permettre l’évaluation de différentes approches thérapeutiques. Boyland et Furth tentent le criblage systématique de substances antitumorales dès la fin des années 1930, mais des difficultés d’approvisionnement en animaux et des problèmes financiers les font abandonner. Lettré en Allemagne et Yoshida au Japon entreprennent un criblage sur la tumeur d’Ehrlich vers la même époque, et Shear au NCI utilise le sarcome 37 pour identifier le principe actif des toxines de Coley (contenant le tumor necrosis factor). Cette dernière tumeur est également utilisée comme modèle de criblage au NCI jusqu’au début des années 1950. Ces travaux de pionniers donnent naissance aux plus récents programmes de criblage de molécules anticancéreuses [45]. Ces premières utilisations de modèles tumoraux pour la détection d’agents anticancéreux ont déjà fait l’objet d’articles détaillés [22, 46].
Utilisation des modèles tumoraux pour le dépistage à grande échelle de produits antitumoraux
En ce qui concerne le précriblage, nous pouvons distinguer deux périodes dans les stratégies de dépistage. De 1940 à 1985, la première étape de criblage est effectuée chez l’animal (en général la souris), et à partir de 1985 celle-ci est précédée d’un précriblage in vitro.
Première période avec précriblage in vivo (1940-1985)
1940-1954
Plusieurs groupes de dépistage se forment aux États-Unis dans les années 1940-1950 (Rhoads, Farber, Gellhorn). D’autres groupes, tels Yoshida à l’université de Tokyo, Haddow à Chester Beatty (Angleterre) et Shear au NCI, poursuivent leurs programmes de dépistage commencés dès les années 1930. À la même époque, l’industrie pharmaceutique fournit à ces différents groupes un grand nombre de composés à tester et de nombreux agents anticancéreux sont découverts.
Bien que les molécules découvertes soient issues de programmes de recherche aux buts très variés *, c’est toujours leur activité antinéoplasique sur des modèles de tumeurs greffées murines qui fut l’élément décisif pour la poursuite de leur évaluation clinique [45]. Par la suite la reconnaissance de l’efficacité de ces molécules sur certains cancers humains sera à l’origine de la mise en œuvre de vastes programmes de dépistage et de développement d’agents anticancéreux utilisant les modèles tumoraux murins [44].

Introduction
Nearly 20 years ago, Schabel noted that in vivo screening was the "cornerstone" of the development of anticancer chemotherapy [33]. We believe that this statement holds true today. Indeed, in the absence of clearly identified, validated targets that would enable antitumour selectivity, in vivo tumour models representing a variety of cancers are still the major tool for the detection and evaluation of new antineoplastic drugs [34] and a necessary step before conducting the first (phase I) clinical studies. Recently, the United States National Cancer Institute (NCI) pointed out that there could be no progress in experimental anticancer chemotherapy without the use of animal experimentation [3]. It should also be born in mind that the activity of the forty or so antineoplastic drugs currently in clinical use were discovered and/or evaluated on murine models.
Tumours transplanted into animals have the advantage of presenting several characteristics of the human disease that they aim to represent. Indeed, tumour-bearing animals possess physiological barriers that restrict the product’s access to the tumour, organs that metabolise and excrete the product, healthy tissues that need to be protected, and metastases, as well as all the rational targets of the cancerous disease itself, including its heterogeneity and its mechanisms of resistance.
Nevertheless, it must be stressed that there are no animal tumour models that unequivocally predict the response to anticancer drugs in man (with the possible exception of L1210 leukaemia for acute lymphoblastic leukaemia) and these models will therefore inevitably give some false positive results. It would be utopian to think that a single model could reflect the behaviour of all human tumours, bearing in mind the diversity of human cancers; each malignant neoplasm being an entity with its own biological behaviour and response to drugs [20], not forgetting the complex interactions that exist between tumour and host and between drug, tumour and host. It is possible that the efficacy of a drug against several animal tumours of the same histological type would predict its efficacy in humans against the same types of tumour with more certainty.
In order to put the current issues surrounding screening for new antitumour products into perspective, we will firstly consider the history of the experimental models used in anticancer chemotherapy, followed by the various current screening strategies, and finally the methodology for evaluating antineoplastic agents in vivo, concentrating on solid tumours.
The history of the development of the earliest tumour models and their use in the screening of anticancer agents
The first animal tumour transplant is attributed to Novinsky (1875), who transferred a nasal tumour and a venereal myxosarcoma in dogs [40]. Around the same time, Wehr published research on the propagation of genital tumours in dogs (1888-1889), and Hanau successfully transferred a vulvar carcinoma in rats [40].
Morau was the first person to conduct a systematic study of the host-tumour relationship, using a murine epithelioma [28]. Later, Jensen published studies on the transplantation of a spontaneous alveolar carcinoma in the white mouse, which are considered as the origin of research into murine tumours in Europe and the United States [25]. Indeed, Jensen’s tumours, as well as other tumours isolated by Ehrlich and Borrel, were successfully obtained and propagated in several laboratories, in particular those of Clowes [10] and Tyzzer [41].
The need to use inbred mice to study cancers was recognised from the early 1900s by many scientists interested in the genetic factors associated with the development of mammary tumours. They contributed to the development of a number of murine strains: C57BL/6, C57BL/10, DBA, BALB/c (Little); C3H, CBA (Strong); AKR (Furth) [29]. It is the development of these inbred mouse strains that permitted the use of transplanted tumours, which are still used today.
The development of methods for screening antitumour substances is intimately linked with the development of animal tumour models. Thus, the development of sarcomas 37 and 180 (mouse), Walker 256 carcinosarcoma (rat), and Ehrlich ascites carcinoma (all developed before 1930) enabled the subsequent evaluation of different therapeutic approaches. Boyland and Furth attempted systematic screening of antitumour substances from the late 1930s, but they had to abandon their work due to difficulties in obtaining the animals and to financial problems. At around the same time, Lettré in Germany and Yoshida in Japan started using Ehrlich tumour for screening, and Shear at the NCI used sarcoma 37 to identify the active substance in Coley’s toxins (containing tumour necrosis factor). The latter tumour was also used as a screening model at the NCI until the early 1950s. These pioneering studies gave rise to the more recent screening programmes for anticancer agents [45]. These early uses of tumour models for detecting anticancer agents have already been discussed in detail elsewhere [22, 46].
The use of tumour models for large-scale screening of antitumour products
As far as prescreening is concerned, the history of screening strategies can be divided into two periods. Between 1940 and 1985, the first stage of screening was carried out in animals (usually in mice), and from 1985 onwards this stage was preceded by in vitro prescreening.
The first period, with in vivo prescreening (1940-1985)
1940-1954
Several screening groups formed in the United States during the period 1940-1950 (Rhoads, Farber, Gellhorn). Other groups, such as Yoshida’s at the university of Tokyo, Haddow’s at Chester Beatty (England) and Shear’s at the NCI, continued screening programmes they had started back in the 1930s. During the same period, the pharmaceutical industry provided these various groups with a large number of compounds to test and many anticancer agents were discovered.
Although the drugs they discovered came from research programmes with very varied aims*, it was always their antineoplastic activity against transplanted murine tumour models that determined whether or not their clinical evaluation would be continued [45]. Later on, when it was recognised that these molecules were effective against certain human cancers, vast programmes were set up to screen for and develop anticancer agents using murine tumour models [44].
